    Abstract
Hepcidin is a peptide hormone that plays an important role in iron metabolism. We have produced a recombinant mouse hepcidin-1 by using baculovirus expression system. Its expression yield was 25 μg/ml when cell culture media were supplemented with a protease inhibitor cocktail. The recombinant mouse hepcidin-1 and synthetic human hepcidin-25 had similar effects on reducing ferroportin expression in J774A cell line and in peritoneal macrophages. However, synthetic human hepcidin-25 was more efficient than recombinant mouse hepcidin-1 in reducing iron concentration in blood circulation (p < 0.01)

    , Article Molecular Informatics ; Volume 32, Issue 8 , 2013 , Pages 742-753 ; 18681743 (ISSN) Jalali Heravi, M ; Mani-Varnosfaderani, A ; Valadkhani, A ; Sharif University of Technology
    2013
    Abstract
A total of 21 833 inhibitors of the central nervous system (CNS) were collected from Binding-database and analyzed using discriminant analysis (DA) techniques. A combination of genetic algorithm and quadratic discriminant analysis (GA-QDA) was proposed as a tool for the classification of molecules based on their therapeutic targets and activities. The results indicated that the one-against-one (OAO) QDA classifiers correctly separate the molecules based on their therapeutic targets and are comparable with support vector machines. These classifiers help in charting the chemical space of the CNS inhibitors and finding specific subspaces occupied by particular classes of molecules. As a next...

    , Article Ecotoxicology and Environmental Safety ; Volume 91 , 2013 , Pages 117-121 ; 01476513 (ISSN) Ghadiri, M ; Kariminia, H. R ; Roosta Azad, R ; Sharif University of Technology
    2013
    Abstract
This paper presents a new method for spectrophotometirc detection of sulfide applying fungal peroxidase immobilized on sodium alginate. The sensing scheme was based on decrease of the absorbance of the orange compound, purpurogallin produced from pyrogallol and H2O2 as substrates, due to the inhibition of peroxidase by sulfide. Absorbance of purpurogallin was detected at 420nm by using a spectrophotometer. The proposed method could successfully detect the sulfide in the concentration range of 0.6-7.0μM with a detection limit of 0.4μM. The kinetic parameters of Michaelis-Menten with and without sulfide were also calculated. Possible inhibition mechanism of peroxidase by sulfide was deduced...

Bench-scaled nano-Fe 0 permeable reactive barrier for nitrate removal

    , Article Ground Water Monitoring and Remediation ; Volume 31, Issue 4 , 2011 , Pages 82-94 ; 10693629 (ISSN) Hosseini, S. M ; Ataie Ashtiani, B ; Kholghi, M ; Sharif University of Technology
    Abstract
There are many fundamental problems with the injection of nano-zero-valent iron (NZVI) particles to create permeable reactive barrier (PRB) treatment zone. Among them the loss of medium porosity or pore blocking over time can be considered which leads to reduction of permeability and bypass of the flow and contaminant plume up-gradient of the PRB. Present study provides a solution for such problems by confining the target zone for injection to the gate in a funnel-and-gate configuration. A laboratory-scale experimental setup is used in this work. In the designed PRB gate, no additional material from porous media exists. NZVI (d 50 = 52 ± 5 nm) particles are synthesized in water mixed with...

    Abstract
Statistically based experimental designs were applied to screen and optimize the bioleaching of spent hydrocracking catalyst by Penicillium simplicissimum. Eleven factors were examined for their significance on bioleaching using a Plackett-Burman factorial design. Four significant variables (pulp density, sucrose, NaNO3, and yeast extract concentrations) were selected for the optimization studies. The combined effect of these variables on metal bioleaching was studied using a central composite design (CCD). Second-order polynomials were established to identify the relationship between the recovery percent of the metals and the four significant variables. The optimal values of the variables...

QSAR modelling of integrin antagonists using enhanced bayesian regularised genetic neural networks

    , Article SAR and QSAR in Environmental Research ; Volume 22, Issue 3-4 , May , 2011 , Pages 293-314 ; 1062936X (ISSN) Jalali Heravi, M ; Mani Varnosfaderani, A ; Sharif University of Technology
    2011
    Abstract
Bayesian regularised genetic neural network (BRGNN) has been used for modelling the inhibition activity of 141 biphenylalanine derivatives as integrin antagonists. Three local pattern search (PS) methods, simulated annealing and threshold acceptance were combined with BRGNN in the form of a hybrid genetic algorithm (HGA). The results obtained revealed that PS is a suitable method for improving the ability of BRGNN to break out from the local minima. The proposed HGA technique is able to retrieve important variables from complex systems and nonlinear search spaces for optimisation. Two models with 8-3-1 artificial neural network (ANN) architectures were developed for describingα 4β 7 and α 4β...

QSAR Modeling of Inhibition Behavior of Diabetes Type II Inhibitors

, M.Sc. Thesis Sharif University of Technology Bigdeli, Arafeh (Author) ; Jalali Heravi, Mehdi (Supervisor)
    Abstract
QSAR studies play an important role in drug design and lead optimization by developing a mathematical relationship between the chemical structures of compounds and their biological activities.By using such an approach one could predict the activities of newly designed compounds before a decision is made whether these compounds should be really synthesized or tested. The main aim of the present work was to develop a QSAR model for predicting the inhibitory activity of acetyl CoA carboxylase (ACC)derivatives as anti-diabetic inhibitors. Since variable selection is a critical step in every QSAR study, four different algorithms, based on Monte Carlo cross-validation techniques, were...

A Study of Ascorbic Acid Electro-Oxidation Process on Silver and Silver Selenide Electrodes

, Ph.D. Dissertation Sharif University of Technology Majari Kasmaee, Laleh (Author) ; Gobal, Fereydoon (Supervisor)
    Abstract
Electrocatalytic oxidation of L-ascorbic acid (L-AA) on silver electrode in alkaline solutions was studied employing cyclic voltammetry, chronoamperometry and impedance spectroscopy techniques. In comparison to Pt, the onset potential for L-AA electro-oxidation process on Ag, in a solution containg 0.02 M L-AA and 0.5 M NaOH, was 0.1 V more negative. Also, L-AA oxidation current increased more than 900% at -0.26 V/SCE. According to voltammety and impedance spectropy findings, although hydroxide anion presence was vital for L-AA oxidation to occur, at potentials more positive than -0.2 V/SCE, hydroxide ion tends to inhibit the oxidation process. Studies on the L-AA oxidation process on Ag in...

, M.Sc. Thesis Sharif University of Technology Sakhaei Nia, Abed (Author) ; Gobal, Freydoon (Supervisor)
    Abstract
Schiff bases are used as prevalent ligands in coordination chemistry and are prepared easily. Their application as mild inhibitory agents for prevention of various steel’s corrosion in acidic solution has been proved. When their structure is altered, multi-purpose application for them is found. This property has led to a motive for research and study. In this study, electronic properties of a group of Schiff bases are investigated. The goal of this study is using quantum mechanics (QM) as a tool for specifying electronic properties of some of these bases. With respect to QM calculation which is done by density functional theory (DFT), their properties as inhibitory agents are studied. First,...

Expression and Purification of Amyloid Beta Recombinant Peptide and the Effecs of Ultrasmall Peptides Enriched in Branch Chain Amino Acids as Inhibitors in Amyloid Formation

, M.Sc. Thesis Sharif University of Technology Rezaei Adariani, Soheila (Author) ; Kalhor, Hamid Reza (Supervisor)
    Abstract
Nowdays, it has been shown that protein misfolding is involved in more than forty diseases including Alzheimer’s and Parkinson’s. Alzheimer’s disease is one of the most common neurodegenerative diseases that disrupts brain function. The main cause of this illness is due to formation of extracellular amyloid plaque from a 42-amino acid-peptide called amyloid beta. Amyloidosis is caused by missfolding of peptides and proteins mainly because of physical and chemical changes in biomolecule or their environment.
Therefore, in this dissertation, using synthetic and recombinant abeta peptide, kinetic of amyloidosis was examined. Ultrasmall peptides (three-amino acid), containing branched chain...

Molecular Dynamics Simulation of Aβ42 Neurotoxicity Inhibition in Order to Alzheimer’s Disease Treatment

, M.Sc. Thesis Sharif University of Technology Khandan, Vahid (Author) ; Firoozabadi, Bahar (Supervisor) ; Saeedi, Mohammad Saeed (Supervisor)
    Abstract
Alzheimer’s disease (AD) is the most common type of dementia in the elderly. The neuropathology and treatment of AD is not precisely determined yet, but according to the pathological studies, AD is associated with presence of toxic soluble oligomers and insoluble senile plaques formed by amyloidosis of Amyloid Beta (Aβ) in neocortical region of brain. An attractive therapeutic approach to treat AD is to identify small ligands that are capable of binding to critical residues in order to inhibit or reverse Aβ amyloidosis process as source of neurotoxicity. In this area, therapeutic efforts designed various organic agents and most of them focused on the N-terminal sequence of Aβ. The aim of...

    Abstract
Abstract In this work, two [60] fullerene derivatives were employed as flow improvers for waxy crude oil to reduce problems caused by the crystallization of paraffin during the production and/or transportation of the crude oil. These amine-based derivatives of [60] fullerene have been applied as wax inhibitor for improvement of rheological performance as well as pour point depressant in a waxy crude oil. Moreover, the influence of these [60] fullerene derivatives on the viscosity and pour point was also investigated in an Iranian waxy crude oil sample. The [60] fullerene derivatives were simply prepared by stirring a solution of [60] fullerene and two lipophilic amines. The results similarly...

    Abstract
The corrosion inhibition effect of zinc acetylacetonate (ZAA) and benzothiazole (BTH) mixture was evaluated for mild steel in 3.5% NaCl solution. To this end, ZAA:BTH mixtures ranged from 6:1 to 1:6 mol ratios were examined by weight loss and open circuit potential to obtain optimal mole ratio. The optimal mixture of ZAA:BTH at 1:5 mol ratio showed a significant corrosion inhibition efficiency proved by electrochemical impedance spectroscopy and polarization studies. The addition of the optimal mixture of ZAA:BTH to epoxy coating showed a considerable increase of corrosion protection evaluated by salt spray exposure
