  7. Abstract:
  8. In recent years, many studies have conducted research about stem cells due to their prominent features such as proliferation and differentiation. By changing the specific parameters of the cellular micro-environment, it is possible to determine the stem cells fate and thereby use them in medical affairs. For this purpose, various devices and methods have been designeds such as microfluidic devices and scaffolds. The microfluidic devices has been especially focuced in the researchs due to their unique features, like the possibility of testing with small amounts of samples, the creation and control of low cost cellular micro-environment and low practical time. Two-dimensional scaffolds made up of micro-fibers and nano-fibers have been widely used in the construction of cell niches due to the high level of flexibility, specific surface, porosity and mechanical strength. Polyvinyl alcohol has been selected as a scaffold in this study. This scaffold has been coated by gold nanoparticles. The microfluidic chip used in this study included two input and output channels and a nanofibre chambern. The main finding in this study is that it is possible to differentiate adult human bone marrow mesenchymal cells to cardiomyocyte cells in a microfluidic chip containing conductive scaffold (to precisely simulate intracellular micro-environment), without differentiation inducing factors. The main reason for this differentiation is the creation of conductive paths for electrical signaling between cells and, consequently, the creation of a beat along the cells. In this study, the differentiation of BMSCs stem cells into cardiomyocytes was evaluated and confirmed by immunocytochemistry, RT-PCR and TEM tests
