    Electrospun polyethersolfone nanofibrous membrane as novel platform for protein immobilization in microfluidic systems

    Abstract
    In the present study, the feasibility of electrospun polyethersolfone (PES) nanofibrous membrane as the solid substrate for microfluidic based immunoassays to enhance the density of immobilized antibody on the surface of membrane was assessed. Conversely, the efficacy of antibody immobilization was compared by two different strategies as 1-Ethyl-3-(3-dimethylaminopropyl)-carbodiimide (EDC)/N-Hydroxysuccinimide (NHS) coupling chemistry and hydrophobic interaction. Compared to conventional immunoassays carried out in plates or gels, microfluidic based immunoassays grant a lot of advantages such as a consumption of little samples and reagents, shorter analysis time, and higher efficiency....

    Abstract
    Background: Hepatitis B virus surface antigen (HBsAg) is an important protein in both diagnosis and prevention of hepatitis B infection. In the current study, a piezoelectric immunosensor based on antibody-antigen interaction was designed to detect HBsAg. A quartz crystal microbalance system was employed to detect antibody-antigen interaction. Methods: At first, an oscillator was designed to measure the resonant frequency affected by the reactants using IC 74LVC1GX04. Antibody against HBsAg was immobilized on 10 MHz AT-cut quartz crystal.     Abstract
    In this paper we introduce novel strategy for antibody immobilization using high surface area electrospun nanofibrous membrane based on ethyl-3-(3-dimethylaminopropyl)-carbodiimide/N-hydroxysuccinimide (EDC/NHS) coupling chemistry. To present the high performance of proposed biosensors, anti-staphylococcus enterotoxin B (anti-SEB) was used as a model to demonstrate the utility of our proposed system.     Abstract
    A label-free electrochemical biosensor was developed for the rapid detection of the matrix metalloproteinase 9 (MMP-9) biomarker on the basis of antibody immobilizing on the zinc oxide (ZnO) nanoparticle and ZnO nanorod electrodes. The charge transfer resistance (Rct) of the electrodes was used as the indicator for MMP-9 concentration, which was obtained through cyclic voltammetry (CV) and electrochemical impedance spectroscopy (EIS). The ZnO nanorod-based biosensor exhibited linear behavior in the MMP-9 concentration range of 1–1000 ng/ml, which is a wider range than the available concentration ranges for most of the conventional methods.     Acute respiratory syndrome coronavirus 2 (SARS-CoV-2), also called COVID-19, is one of the most contagious viruses resulting in a progressive pandemic.
